Abstract
[fr] Ce travail a pour but de résumer les principaux points du système immunitaire inné interférant avec la mise en place d’une réponse immunitaire suite à une vaccination ARN, ainsi que les méthodes de contournement couramment utilisées.
La production in vitro d’ARNm, réalisée grâce à la transcription d’un ADN cible, permet l’ouverture vers une nouvelle voie de vaccination. Cependant, lors de l’injection d’un ARN exogène dans un organisme vivant, de nombreux mécanismes de défense contre cette chaine nucléotidique se mettent en place. On retrouve notamment la reconnaissance de cet ARN par les PRR (comme RIG, TLR 7, TLR 8, …). Une fois activé, le système immunitaire inné va détériorer l’ARNm et rendre son action impossible. Afin d’y pallier, bon nombre de mécanismes agissant sur les différentes étapes importantes de reconnaissance par le système immunitaire inné se sont développés.
Premièrement, une étape de purification post IVT est réalisée. Cette étape permet une purification de l’ARN simple brin en le séparant des ARN double brins présents dans l’échantillon.
La deuxième étape consiste en l’ajout d’une coiffe et d’une queue polyA. Le but de ces deux techniques est d’éviter la destruction de l’ARNm par les ARNases tout en régulant l’expression des protéines traduites à partir de l’ARN d’intérêt. D’autres étapes de modification de l’ARN sont également décrites, telle que la modification des nucléotides ou des bases.
Le transport de ces molécules d’ARNm, vers l’endosome d’une cellule, représente également une étape primordiale. Pour ce faire, deux grands concepts ont vu le jour. Le premier, et le plus largement utilisé, comprend l’utilisation de lipides chargés positivement liant l’ARN et le protégeant de la reconnaissance des acteurs du système immunitaire.
La deuxième méthode de transport quant à elle, fait appel à un copolymère amphiphile.
On constate à travers ces différentes techniques, une progression importante de la technologie ARN. Il est désormais possible de contourner de manière efficace le système immunitaire d’un individu. A l’avenir, d’autres domaines médicaux, dont certains déjà très développés, pourraient également bénéficier de cette nouvelle approche basée sur l’utilisation d’ARNm.
[en] The aim of this work is to summarize the main points of the innate immune system interfering with the establishment of an immune response following RNA vaccination, as well as the bypassing methods commonly used.
The in vitro production of mRNA, achieved through the transcription of a target DNA, allows the opening of a new vaccination route. However, when an exogenous RNA is injected into a living organism, many defense mechanisms against this nucleotide chain are put in place. In particular, this RNA is recognized by PRRs (such as RIG, TLR7, TLR8, ...). Once activated, the innate immune system will damage the mRNA and make its action impossible. In order to overcome this, a number of mechanisms acting on the different important steps of recognition by the innate immune system have been developed.
First, a post IVT purification step is performed. This step allows a purification of the single-stranded RNA by separating it from the double-stranded RNA present in the sample.
The second step consists in the addition of a cap and a polyA tail. The goal of these two techniques is to avoid the destruction of the mRNA by RNAases while regulating the expression of the proteins translated from the RNA of interest. Other RNA modification steps are also described, such as nucleotide or base modification.
The transport of these mRNA molecules to the endosome of a cell is also an important step. For this purpose, two main concepts have emerged. The first, and most widely used, involves the use of positively charged lipids that bind the RNA and protect it from recognition by the immune system.
The second transport method uses an amphiphilic copolymer.
Through these different techniques, we can see an important progression of the RNA technology. It is now possible to effectively bypass the immune system of an individual. In the future, other medical fields, some of which are already highly developed, could also benefit from this new approach based on the use of mRNA.
