Abstract

[en] Nitrogen-containing heterocycles find various applications in pharmaceutical chemistry, ranging from antibiotics to antihistamines, highlighting their importance. Among them, 1,2-oxazine rings arise as a promising motif for both synthetic applications, through their high functionalization potential, as well as in drug discovery due to their polar nitrogen-oxygen bond, allowing interactions with a wide variety of biological receptors. Various methods have been developed to access 1,2-oxazines, one of the main approaches relies on the generation of nitrosocarbonyls from bench-stable precursors and their subsequent Diels-Alder reaction with 1,3-diene systems. This method is highly advantageous because of the high functionalization that can be introduced through the nitrosocarbonyl precursor. Nonetheless, the transient nature of nitrosocarbonyl species makes their reactivity hard to harness. As a result, this thesis aims to explore novel and controllable strategies for the generation of 1,2-oxazine rings using nitrosocarbonyls.

Firstly, the assessment of flow photochemical oxidation conditions for the generation of nitrosocarbonyls was performed. Using organic dyes as photosensitizers to generate singlet oxygen, the aim is to develop a traceless catalytic method. However, selectivity limitations were quickly identified due to competing diene oxidation. Nevertheless, triplet oxygen was found to efficiently generate the nitrosocarbonyl, leading to oxazine formation. Experimental mechanistic insights were supported by DFT and CCSD(T) computational studies.

Secondly, a TEMPO-mediated oxidation of nitrosocarbonyl precursors is described. Inspired by TEMPO catalyzed alcohol oxidation, tert-butyl hypochlorite was introduced as a secondary oxidant. Encouraged by initial results, an optimization of reaction conditions was conducted, followed by kinetics studies to investigate the mechanistic pathway associated with this protocol.

[fr] Les hétérocycles contenant de l’azote trouvent de nombreuses applications en chimie pharmaceutique, allant des antibiotiques aux antihistaminiques, soulignant ainsi leur importance. Parmi eux, les cycles 1,2-oxazine apparaissent comme un motif prometteur, tant pour des applications synthétiques, en raison de leur fort potentiel de fonctionnalisation, que pour la découverte de médicaments, grâce à la polarité de la liaison azote–oxygène, qui permet des interactions avec une grande variété de récepteurs biologiques. Différentes méthodes ont été développées pour accéder aux 1,2-oxazines, l’une des principales approches repose sur la génération de nitrosocarbonyles à partir de précurseurs stables, suivie de leur réaction de Diels–Alder avec des systèmes 1,3-diènes. Cette méthode est particulièrement avantageuse en raison du haut degré de fonctionnalisation pouvant être introduit via le précurseur nitrosocarbonyle. Néanmoins, le caractère transitoire des espèces nitrosocarbonyles rend leur réactivité difficile à exploiter. Dans ce contexte, ce mémoire vise à explorer des stratégies nouvelles et contrôlables pour la génération de cycles 1,2-oxazine à partir de nitrosocarbonyles.

Dans un premier temps, l’évaluation de conditions d’oxydation photochimique en flux pour la génération de nitrosocarbonyles a été réalisée. L’utilisation de colorants organiques comme photosensibilisateurs pour générer de l’oxygène singulet visait le développement d’une méthode catalytique et sans trace. Toutefois, des limitations de sélectivité ont rapidement été mises en évidence en raison de l’oxydation concurrente du diène. Néanmoins, l’oxygène à l’état triplet s’est révélé capable de générer efficacement le nitrosocarbonyle, conduisant à la formation de l’oxazine. Des analyses mécanistiques expérimentales ont été corroborées par des études computationnelles de type DFT et CCSD(T).

Dans un second temps, une oxydation médiée par TEMPO des précurseurs nitrosocarbonyles est présentée. Inspirée de l’oxydation des alcools catalysée par TEMPO, l’hypochlorite de tert-butyle a été introduit comme oxydant secondaire. Encouragé par les résultats initiaux, une optimisation des conditions réactionnelles a été menée, suivie d’études cinétiques visant à élucider le chemin réactionnel associé à ce protocole.
