Abstract

[en] This work is a part of a project funded by the U.S. Food and Drug Administration (FDA) and aims at the synthesis of active pharmaceutical ingredients (APIs) from renewable building blocks. Three essential drugs were selected as case studies: Timolol, Indomethacin, and Nitrofurantoin. The present work focuses on Timolol, which can be prepared from glycerol, a highly promising chemical derived from biomass. Glycerol can serve as a precursor for a variety of valuable chemicals, particularly glycidol, which constitutes the starting point of this study.
Three synthetic pathways sharing a common initial step were investigated. This first step consists of the aminolysis of glycidol, developed and optimized under continuous flow conditions. Subsequently, three different strategies were explored, starting from the same intermediate synthesized in the initial common step. The first pathway involves the protection of the aminolysis product using dimethyl carbonate (DMC). The second pathway relies on the direct reaction of the aminolysis product with 4-(4-chloro-1,2,5-thiadiazol-3-yl)morpholine without any protection step. The third pathway involves the protection of the aminolysis product using benzaldehyde. All three pathways were evaluated with the objective of designing an environmentally friendly and sustainable synthetic route to Timolol.

[fr] Ce travail s’inscrit dans le cadre d’un projet financé par la Food and Drug Administration (FDA) visant à développer des voies de synthèse d’ingrédients pharmaceutiques actifs (APIs) à partir de briques moléculaires renouvelables. Trois médicaments essentiels ont été retenus comme cas d’étude : le Timolol, l’Indométhacine et le Nitrofurantoïne. Le travail suivant se concentre plus particulièrement sur le Timolol, dont la synthèse peut être envisagée à partir de glycérol, une plateforme chimique d’origine biomasse particulièrement prometteuse. Le glycérol constitue en effet un précurseur de nombreux composés à haute valeur ajoutée, notamment le glycidol, qui représente le point de départ de cette étude.
Trois voies de synthèses distinctes, partageant une étape initiale commune, ont été explorées. Cette première étape correspond à la réaction d’aminolyse du glycidol, développée et optimisée en conditions de flux continu. Les différentes stratégies ont ensuite été mise en œuvre à partir d’un même intermédiaire synthétisé lors de cette étape initiale commune. La première voie repose sur la protection du produit de l’aminolyse à l’aide du carbonate de diméthyle. La deuxième voie consiste en la réaction directe du produit d’aminolyse avec la 4-(4-chloro-1,2,5-thiadiazol-3-yl)morpholine, sans étape de protection préalable. Enfin, la troisième voie implique la protection du produit d’aminolyse à l’aide du benzaldéhyde. L’ensemble de ces voies a été étudié dans une optique de conception d’une synthèse du Timolol plus respectueuse de l’environnement et durable.
